I am using RFSoc 4x2 board to design a multi-tone generator using the DAC ports. Is this possible to generate multiple frequencies using one single DAC port?

Yes this is entirely possible. One method to do this is to create multiple tones at baseband then use the NCO to mix them up to RF.
